چکیده

a-Isopropylmalate synthase, the first specific enzyme in leucine biosynthesis, was purified approximately 100-fold from extracts of Saccharomyces sp. (strain 60615), the most effective step being specific elution with the feedback inhibitor leucine from a hydroxyapatite column. In the early steps of purification, special care was taken to protect the synthase against proteolytic activities. The apparent molecular weight of the enzyme as determined from gel filtration

CLONING AND CHARACTERIZATION O F YEAST LEU 4 , ONE O F T W O GENES RESPONSIBLE FOR a - ISOPROPYLMALATE SYNTHESIS

By complementation of an a-isopropylmalate synthase-negative mutant of Saccharomyces cerevisiae (leu4 leu3), a plasmid was isolated that carried a structural gene for a-isopropylmalate synthase. Restriction mapping and subcloning showed that sequences sufficient for complementation of the leu4 leu5 strain were located within a 2.2-kilobase S a l I h I I segment. Southern transfer hybridization ...

Kinetics Studies Impact of Initial pH and Addition of Yeast Saccharomyces cerevisiae on Biogas Production from Tofu Wastewater in Indonesia

The purpose of this work was to study the effect of initial pH and yeast Saccharomyces Cerevisiae on biogas production from tofu wastewater (TW). The initial pH was varied in ranging of 5 – 9 in substrate without yeast (T5-T9) and with yeast (TY5-TY9). The results showed that optimum initial pH was 8. The maximum biogas was resulted in T8 (275 mL) and TY8 (421 mL). Yeast addition increased tota...

Evidence for Two Distinct CoA Binding Sites on Yeast cy- Isopropylmalate Synthase*

cu-Isopropylmalate synthase (EC 4.1.3.12) from Saccharomyces cerevisiae was purified to a purity of about 95%. The molecular weight of the enzyme is approximately 127,000, as determined by sedimentation equilibrium centrifugation and by intersubunit cross-linking. Under denaturing conditions, one major species (95%) with molecular weight of about 65,000 is obtained. The dimeric structure of the...

Purification of yeast alpha-isopropylmalate isomerase. High ionic strength hydrophobic chromatography.

alpha-Isopropylmalate isomerase, the second enzyme specific for leucine biosynthesis, can be purified from extracts of yeast utilizing a chromatographic procedure that allows separation of proteins in the presence of high concentrations of (NH4)2SO4. The purification procedure utilizes the stabilizing effect of glycerol and (NH4)2SO4 on the isomerase and their opposing effects on protein retent...

Yeast alpha-isopropylmalate isomerase. Factors affecting stability and enzyme activity.

Yeast alpha-isopropylmalate isomerase was found to be markedly stabilized by high concentrations of glycerol and (NH4)2SO4. Such conditions of high ionic strength inhibited the enzyme, stabilized the enzyme to heat, and affected kinetic parameters. The isomerase was found to exhibit ionic strength-dependent hysteresis when enzyme, totally but reversibly inhibited by storage under conditions of ...
